Could anyone steer me in the right direction on where the fuse box is on my 75 250 6 cyl. I cant find it and dont have a owner or shop manual .. Have no dome lights or front side markers... Do those front side markers work as side signals ??

Dome light and side markers are on a different circuit. Fuse box is on the firewall...very top-left side

No power now to nothing att all..
Is this screw with spring a ground wire ?? What the heck have I done ??

You won't get power to anything until you re-install the fuse box. That's grand-central station for power in your car. Hope you disconnected the battery negative terminal first, because you just pulled the main disconnect with live hot wires when you separated the bulkhead connector. Never know what you might fry, including yourself on the two fuse holders that are always hot. Specifically, the bolt holds the two halves of the bulkhead connector together, plus the fusebox since it is attached to the inside half. Power from the battery goes through there to the ignition switch, and all power in and out of the engine compartment goes through that connector (with a couple minor exceptions).
